In the webinar, we would introduce a new UCL facility that provides nucleic acid synthesis and molecular cloning services. The aim will be to facilitate synthetic biology processes for UCL scientists to bring these usually outsourced technologies in-house for the first time.

About the new facility

The facility will be based in Charles Bell House research laboratories in Fitzovia, led by Dr John Counsell’s research group. Users will be able to request a range of molecular biology services using the newly installed BioXp platform, which delivers automated, overnight DNA, mRNA, or peptide synthesis.

Services include:

• De novo synthesis of DNA fragments up to 7 kb with optional cloning into custom plasmid

• De novo synthesis of scanning libraries up to 3kb

• De novo mRNA synthesis up to 1.8kb (clean-cap, unmodified or pseudouridine-modified)

• In vitro cell-free DNA plasmid amplification

• In vitro mRNA synthesis up to 10kb from starter plasmid

Potential applications of the technology in research and teaching include:

• Synthesis of custom DNA or mRNA fragments for molecular biology

• Automated cloning of genes into gene therapy plasmids

• Discovery of novel protein variants through scanning library synthesis

• Engineering and discovery of novel antibodies

• Cell-free DNA and mRNA amplification (without the use of GMOs)
